The new STM32G474RE has an ADC that can run up to 4MSPS.
I'm doing a project where I need to do the sampling in 1MSPS. But the maximum sample rate achieved was 300kSPS. When I set timer 1 for the ADC trigger to 500kSPS the "while" loop simply locks. Seems to be all right with ADC setup, but I can't solve this problem.

Looks like HAL needs faster processor to keep up with the ADC.
HAL is not code or speed efficient

The processor is running at 160MHz and the ADC uses reading via DMA. Do you refer to the ADC lib HAL or the other peripherals?
Even using the HAL and DMA libs, I am still a long way from 4MSPS, and that I cannot understand.
If I start reading 4 channels in 250KSPS the same code works very well, and in my understanding it is the equivalent of reading 1 channel in 1MSPS.

I don't use Cube, but doesn't this mean that the DMA ISR will be called after every single ADC conversion?

And the interrupt service takes longer then a single ADC conversion.

I can't see any interrupt handler code.
Check it's runtime, including all the HAL-generated callbacks.
I don't use Cube either.

I managed to solve the problems.
HAL actually enables all possible interruptions. The solution was to disable all ADC and DMA interruptions:

Later I only enabled DMA TC interrupt:

This way I got 500KSPS.
Finally I changed the function to clear the interrupt flag (HAL_DMA_IRQHandler(&hdma_adc1)) to write directly to the register IFCR:
