Well reviewing the CM3 TRM, or one of Joseph Yiu's Essential series books will provide context for the ST Reference Manual.

I'll note I came here proficient in C and ARM Assembler. If you have familiarity with other micro-controllers the path will be easier, the CM3/CM4 are an evolution over the earlier ARM cores.
