I am trying to periodically wakeup STM32L010K8 from standby mode. The project is generated is using CubeMX. The device doesn't wakeup unless I reinitialize RTC everytime it resumes from standby. How can I avoid reinitialization of RTC everytime?

You clear the standby flag on a reset but I don't see where you clear the RTC event/interrupt that caused the wakeup. A wakeup after standby requires an edge transition on the event or interrupt, so if you don't clear it first there won't be a transition the second time you enter standby. Resetting RTC works because you clear the pending event when you configure the RTC.

I have thought using the following is enough.
__HAL_PWR_CLEAR_FLAG(PWR_FLAG_WU);
The following worked for me:
hrtc.Instance = RTC;
__HAL_RTC_WAKEUPTIMER_CLEAR_FLAG(&hrtc, RTC_FLAG_WUTF);
__HAL_PWR_CLEAR_FLAG(PWR_FLAG_WU);
HAL_PWR_EnterSTANDBYMode();
