SPI hardware Design consideration

Hello 

In my board there are two uc that are connecting through SPI .Is it necessary to have pullup resistor for spi pins ?

It is certainly advisable on at least the Chip Select / Slave Select line or lines.

That said, you can generally configure STM32 I/O lines for internal pullup/pulldown.
